腸道功能研究是21世紀最熱門的研究領域之一!

  本實驗室成立於2017年夏天。歡迎本系大學部與研究所同學、醫學院營養相關科系或生命科學相關科系背景有志於腸道免疫與營養探索與研究之同學或專業人士加入本研究室團隊。
實際研究貢獻成果皆會列名發表於相關領域研討會與國際期刊。

研究簡介

自2007與2013年美國國家衛生研究院(National Institutes of Health, NIH)進行二階段「人體微生物組研究計畫(Human Microbiome Project)」,腸道功能即成為當今科學界之熱門研究標的。
 

  腸粘膜系統 (Intestinal mucosa), 主要由腸道上皮細胞 (intestinal epithelium)與固有層內的腸淋巴組織(gut-associated lymphoid tissue, GALT )之免疫細胞所組成,是人體消化道區隔外在物質與內在環境的第一屏障
  覆蓋於人體消化系統之上,是一單層上皮細胞是區隔身體內在環境與消化管腔外在環境之介面。除了營養素的消化與吸收之外,在空間上,細胞頂面(apical side)面對腸道微生物; 底面 (basolateral side)存在大量免疫細胞與神經叢,其肩負了雙向溝通之角色。位在腸組織腺窩(crypt)底部之腸幹細胞(intestinal stem cell),規律地進行增生並分化成不同功能之腸上皮細胞,包括吸收型腸細胞 (absorptive enterocytes)、杯狀細胞 (goblet cells)、神經內分泌細胞 (neuroendocrine cells)、tuft cells、Paneth cells 與 M cells;而移動至頂端老舊細胞每3-5天發生凋亡。在新生與凋亡之間遞補更迭,形成巧妙之恆定。

研究方向:

  消化道之小腸是人體攝取生命所需養份的第一道關卡,大腸則是將攝入養分去蕪存菁之後排出體外的中繼站。腸道上皮細胞、腸道管腔物質(微生物與營養素)、腸免疫細胞之三相溝通與代謝反應,以及Gut-brain axis (腦腸軸)之溝通,構築與形塑人體各部機能之恆定。本實驗室之研究目標,主要為探討單一營養素或飲食型態組成,在生理與病理狀態中,於腸道微觀與巨觀之角色。

實驗室技術

  大小鼠手術 (大鼠動靜脈氣管插管、腸道手術、心、肺、腎手術模式)、動物缺血模式、細胞株培養、肝臟初代細胞培養、腸道類器官培養、免疫染色技術、分生實驗、非致病性微生物培養... 等

About the Lab
  Dr. Huang’s laboratory started up in the summer of 2017. Her research interests are in the area of gastrointestinal physiology, mucosal immunology and nutrition. Utilizing the animal and cell culture model, she investigated the role of a single nutrient and mixed diet under physiological and pathological conditions. The techniques she used includes animal surgery, cell culture, intestinal organoid culture, non-pathogenic bacterial culture, immunoassay, histology stating and DNA & RNA extraction and analysis.
